{"id":2457,"date":"2025-08-13T14:45:53","date_gmt":"2025-08-13T14:45:53","guid":{"rendered":"https:\/\/en.ige.uobaghdad.edu.iq\/?p=2457"},"modified":"2025-08-21T01:03:57","modified_gmt":"2025-08-21T01:03:57","slug":"genetic-engineering-discusses-the-molecular-detection-and-gene-expression-of-efflux-pump-genes-and-virulence-factors-in-pseudomonas-aeruginosa-isolated-from-clinical-sources-ask-chatgpt","status":"publish","type":"post","link":"https:\/\/en.ige.uobaghdad.edu.iq\/?p=2457","title":{"rendered":"Genetic Engineering Discusses the Molecular Detection and Gene Expression of Efflux Pump Genes and Virulence Factors in Pseudomonas aeruginosa Isolated from Clinical Sources"},"content":{"rendered":"<div class=\"wpb-content-wrapper\"><p>[vc_row][vc_column][vc_column_text]<\/p>\n<p style=\"text-align: justify;\" data-start=\"0\" data-end=\"184\"><span style=\"font-size: 18px;\">The Institute of Genetic Engineering and Biotechnology for Postgraduate Studies at the University of Baghdad discussed the master\u2019s thesis of student Sameer Adel Jawad, entitled:<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"186\" data-end=\"390\"><span style=\"font-size: 18px;\">\u201cMolecular Detection and Gene Expression of Efflux Pump Genes and Virulence Factors in <em data-start=\"275\" data-end=\"299\">Pseudomonas aeruginosa<\/em> Isolated from Clinical Sources\u201d,<\/span><br data-start=\"334\" data-end=\"337\" \/><span style=\"font-size: 18px;\">supervised by Professor Wathiq Abbas Al-Draghi.<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"392\" data-end=\"511\"><span style=\"font-size: 18px;\">The study aimed to highlight the widespread prevalence of resistance and virulence determinants in clinical isolates.<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"513\" data-end=\"880\"><span style=\"font-size: 18px;\">Using quantitative real-time PCR (qRT-PCR) with the <em data-start=\"565\" data-end=\"570\">fbp<\/em> gene as an internal housekeeping reference, the study revealed distinct gene expression patterns across different resistance categories. Sensitive isolates showed relatively low expression levels of both virulence genes and efflux pump\u2013related genes (<em data-start=\"822\" data-end=\"828\">ExoS<\/em>: 1.18, <em data-start=\"836\" data-end=\"842\">ToxA<\/em>: 0.45, <em data-start=\"850\" data-end=\"856\">MexA<\/em>: 1.59, <em data-start=\"864\" data-end=\"870\">MexB<\/em>: 3.64).<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"882\" data-end=\"1100\"><span style=\"font-size: 18px;\">In contrast, multidrug-resistant (MDR) isolates demonstrated a marked increase in the expression of efflux pump genes (<em data-start=\"1001\" data-end=\"1007\">MexB<\/em>: 6.91, <em data-start=\"1015\" data-end=\"1021\">MexA<\/em>: 2.87, <em data-start=\"1029\" data-end=\"1035\">OprM<\/em>: 1.45), accompanied by moderate expression of virulence genes.<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"1102\" data-end=\"1324\"><span style=\"font-size: 18px;\">The qRT-PCR analysis showed clear sigmoidal amplification curves for all target genes, indicating active transcription. Gene expression was quantified using the \u0394\u0394CT method, and fold changes were calculated as 2^(-\u0394\u0394CT).<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"1326\" data-end=\"1467\"><span style=\"font-size: 18px;\">The study concluded that the widespread presence of resistance and virulence genes in these clinical isolates has significant implications.<\/span><\/p>\n<p style=\"text-align: justify;\" data-start=\"1469\" data-end=\"1644\" data-is-last-node=\"\" data-is-only-node=\"\"><span style=\"font-size: 18px;\">It recommended that the pervasive occurrence of the <em data-start=\"1521\" data-end=\"1533\">MexAB-OprM<\/em> efflux pump genes indicates a high likelihood of antibiotic resistance, which could lead to treatment failure.<\/span><\/p>\n<p>[\/vc_column_text][\/vc_column][\/vc_row][vc_row][vc_column width=&#8221;1\/2&#8243;][vc_single_image image=&#8221;2454&#8243; img_size=&#8221;large&#8221; alignment=&#8221;center&#8221; onclick=&#8221;img_link_large&#8221; css=&#8221;&#8221;][\/vc_column][vc_column width=&#8221;1\/2&#8243;][vc_single_image image=&#8221;2453&#8243; img_size=&#8221;large&#8221; alignment=&#8221;center&#8221; onclick=&#8221;img_link_large&#8221; css=&#8221;&#8221;][\/vc_column][\/vc_row]<\/p>\n<\/div>","protected":false},"excerpt":{"rendered":"<p>[vc_row][vc_column][vc_column_text] The Institute of Genetic Engineering and Biotechnology for Postgraduate Studies at the University of Baghdad discussed the master\u2019s thesis &#8230; <\/p>\n","protected":false},"author":37,"featured_media":2455,"comment_status":"closed","ping_status":"open","sticky":false,"template":"","format":"standard","meta":{"_lmt_disableupdate":"no","_lmt_disable":"","footnotes":""},"categories":[14,17,7],"tags":[],"class_list":["post-2457","post","type-post","status-publish","format-standard","has-post-thumbnail","hentry","category-archives","category-last-news","category-a-a"],"yoast_head":"<!-- This site is optimized with the Yoast SEO plugin v25.8 -
